Not unless you intentionally ride on tacks. Tubeless tires are very durable, don't get pinch flats, and are often filled with that gel stuff that seals punctures for you.I agree with Angus. I've found that even when a tubeless tire gets punctured, it "leaks" rather than "pops" as a tube does. I'll take a leak over a pop any day! Add some Stan's or other tire sealant to the equation and you almost completely negate the need to remove the tire from the rim..except to replace it.
