I'm searching for a waterproof duffle I can strap to the back of a motorcycle that is Waterproof. This bag says its waterproof, but with an exposed zipper (even with a flap),....how can it be as waterproof as say a Rolltop? Has anyone really tested this bag in a downpour for hours at a time with the wind blowing across the top at 70mph?

Look at the SealLine website. This is MORE waterproof then a roll-top. Rated for 1meter for 30 mins.

I just use a regular dry bag on my motorcycle. I was off road going through streams nothing got wet.

Not on the back of a motorcycle, but a friend of mine has one, and he brought it with us on a trip to Jasper a little while back. It was in the box of the truck, pouring rain going about 110 km/h and was completely dry to the touch on the outside as soon as we were out of the storm, and was bone dry inside when we arrived.
