You cannot lock the bikes, you would need to buy a lock, but you can secure them with the straps provided.

I have a spare tire mounted above my hitch on both vehicles I would use this rack on. Question is there enough clearance for the rack or will I have to buy an extender?

Some friends of mine have a Toyota RAV4 and their rack works just fine. Check out the image to see if the style is similar to your vehicles.

Are you dead set on hitch rack in this style? Bikes swing back and forth and collide with one another, and if you have any dual suspension bikes you're just asking for mounting troubles.
Personally I prefer fork mount roof racks, but I'm a huge fan of Yakima's tray racks. They're quite a bit more expensive, but I like to think that my bike deserves the best after I ride it so hard.
