Yes, I have packed a Canon dSLR and lens in a LowePro soft case, canon 70-250 lens, tripod, flash, batteries, along with a jacket, maps, small first aid kit, and food for a day, while still being able to fill the bladder. It feels a bit stretched out when packed to the brim, but it still fits comfortably.
However, this is not designed to be a camera backpack. LowePro has some excellent options that will make accessing your camera easier, as well as storing it safer.
Will this be comfortable for a 4-5 hour hike? I'm looking to put in a DSLR sized camera, some food, small gear (handheld gps, flashlights, knife, etc.), and maybe a sweater.
For those that have had this, how does it hold up to rain? Camelbak says it has a waterproof main compartment. I can't find many packs in this size range that would fit my need to protect my camera from water. Considered the Vantage 40, but that seems too big for my needs, and the H.A.W.G. NV seems too small. If anyone has any other suggestions, I'm all ears.
The vantage 40 would actually be a good choice if your camera takes up a lot of room (and given your other listed gear). The vantage 40 would also allow you to have more options for carrying gear on the outside of your pack.

This is a roll-top closure, this you can tell from the picture. I have a Camelbak Vantage 40 with the same roll-top closure. Then it has Velcro, buckles and fasteners. However, it is not 100% submersible, more functional for having rain stay out. You could take it out on the water but it will not replace a dry bag with the same roll-top closure.
I too am confused about this... The camelbak website says that it has a waterproof main compartment. I'm looking for something that will keep my camera dry with enough storage space for a long day hike. Originally I looked into the HAWG NV, but now I'm thinking that it's too small, so the waterproof thing is a big deal with me.
